Ion Exchange Chromatography | Principle, Instrumentation & Lab Experiment
This video lecture talks about Ion exchange chromatography in Hindi, Ion Exchange chromatography, cation exchange chromatography, anion exchange chromatography, ion exchange chromatography practicle, lab procedure, PI of Protein, Zwitter ion, Ph of buffer solution in ion exchange chromatography. Ion exchange chromatogram This lecture explains about the instrumentation, principle of Ion exchange Chromatography. Ion chromatography is a chromatography process that separates ions and polar molecules based on their affinity to the ion exchanger. It works on almost any kind of charged molecule—including large proteins, small nucleotides, and amino acids. However, ion chromatography must be done in conditions that are one unit away from the isoelectric point of a protein. The two types of ion chromatography are anion-exchange and cation-exchange. Cation-exchange chromatography is used when the molecule of interest is positively charged. The molecule is positively charged because the pH for chromatography is less than the pI. In this type of chromatography, the stationary phase is negatively charged and positively charged molecules are loaded to be attracted to it. Anion-exchange chromatography is when the stationary phase is positively charged and negatively charged molecules (meaning that pH for chromatography is greater than the pI) are loaded to be attracted to it.It is often used in protein purification, water analysis, and quality control. The water-soluble and charged molecules such as proteins, amino acids, and peptides bind to moieties which are oppositely charged by forming ionic bonds to the insoluble stationary phase. The equilibrated stationary phase consists of an ionizable functional group where the targeted molecules of a mixture to be separated and quantified can bind while passing through the column—a cationic stationary phase is used to separate anions and an anionic stationary phase is used to separate cations. Cation exchange chromatography is used when the desired molecules to separate are cations and anion exchange chromatography is used to separate anions. Ion Exchange Chromatography
The principle of Ion exchange chromatography is based on the attraction of oppositely charged particle. Many of the biological compounds such as protein, amino acid have charges. These charges can be used in the separation of the compounds by using ion exchanger. The Exchangers are Anionic exchanger which are positively charged and Cationic exchanger which are negatively charged.

Chromatography 101: An Introduction to Ion Exchange Chromatography
For more information, visit http://www.bio-rad.com/yt/26/ngc. Shawn Anderson presents an introduction to ion exchange chromatography using Bio-Rad's chromatography instruments, columns, and media to demonstrate the technique. Shawn covers the basic principles of ion exchange chromatography and touches on applications for the separation of proteins and other biomolecules using a variety of anion and cation exchange media. Bio-Rad's Successful Chromatography Webinar series provides a great introduction to the different chromatography methods available for research applications. Most medium pressure chromatography columns work with Bio-Rad's NGC preparative chromatography system. The NGC is designed to rapidly automate the purification of biomolecules. The flexible, modular, and economical design makes the NGC systems the instruments of choice for method development and scale-up. NGC systems are available in six preplumbed, factory-tested configurations with two different flow rates. Each preconfigured system can be further customized and upgraded by adding valves, detectors, or pumps to meet specific application needs. Any system can be configured for operation at either high or low flow rate by simply changing the pump head on the system pump module. With the NGC system, a single hardware platform can be modified as needed to suit changing applications and scales. . http://www.bio-rad.com/evportal/destination/product?catID=MFCV7415&WT.mc_id=yt-pfd-ww-ngc-20130701-zytDmnoioJo We Are Bio-Rad. Demineralised - De Ionised Mixbed Ion Exchange Water Plant in Pakistan
http://www.pakwatercare.org Deionizers -- Demineraliser Pakwater Care Services is a leading provider of deionization solutions. Our water deionizers are rugged, pre-engineered, pre-assembled, standardized units that minimize expensive installation and start-up costs. We have designed our Deionization systems to maximize the efficiency and repeatability of the unit during the service and regeneration modes The Process of Deionization or Ion-Exchange In the context of water purification, ion-exchange is a rapid and reversible process in which impurity ions present in the water are replaced by ions released by an ion-exchange resin. The impurity ions are taken up by the resin, which must be periodically regenerated to restore it to the original ionic form. (An ion is an atom or group of atoms with an electric charge. Positively-charged ions are called cations and are usually metals; negatively-charged ions are called anions and are usually non-metals). The following ions are widely found in Raw Waters: Cations Anions Calcium (Ca2+) Chloride (Cl-) Magnesium (Mg2+) Bicarbonate (HCO3-) Sodium (Na+) Nitrate (NO3-) Potassium (K+) Carbonate (CO32-) Iron (Fe2+) Sulfate (SO42-) Ion Exchange Resins There are two basic types of resin -- cation-exchange and anion-exchange resins. Cation exchange resins will release Hydrogen (H+) ions or other positively charged ions in exchange for impurity cations present in the water. Anion exchange resins will release hydroxyl (OH-) ions or other negatively charged ions in exchange for impurity anions present in the water. The application of ion-exchange to Water Treatment and Purification There are three ways in which ion-exchange technology can be used in water treatment and purification: first, cation-exchange resins alone can be employed to soften water by base exchange; secondly, anion-exchange resins alone can be used for organic scavenging or nitrate removal; and thirdly, combinations of cation-exchange and anion-exchange resins can be used to remove virtually all the ionic impurities present in the feedwater, a process known as deionization. Water deionizers purification process results in water of exceptionally high quality. Deionization For many laboratory and industrial applications, high-purity water which is essentially free from ionic contaminants is required. Water of this quality can be produced by deionization. The two most common types of deionization are: * Dual-Bed Deionization * Mixed-Bed Deionization Dual-Bed Deionization The two-bed deionizer consists of two vessels -- one containing a cation-exchange resin in the hydrogen (H+) form and the other containing an anion resin in the hydroxyl (OH-) form. Water flows through the cation column, whereupon all the cations are exchanged for hydrogen ions.To keep the water electrically balanced, for every monovalent cation, e.g. Na+, one hydrogen ion is exchanged and for every divalent cation, e.g. Ca2+, or Mg2+, two hydrogen ions are exchanged. The same principle applies when considering anion-exchange. The decationised water then flows through the anion column. This time, all the negatively charged ions are exchanged for hydroxide ions which then combine with the hydrogen ions to form water (H2O). Cation exchange chromatography
Ion exchange chromatography can be done using either an anion or a cation exchanger. When we have to separate cations from a mixture of anions, and neutral molecules, then we should use cation exchange chromatography. Oligosaccharide Analysis: High-Performance Anion-Exchange Chromatography
Oligosaccharide profiling of biopharmaceutical and other glycoprotein products remains challenging for scientists who report that regardless of the technique used, more than one technique is often required for complete oligosaccharide identification. Recent collaborative studies have noted that while MALDITOF MS was faster, High-Performance Anion-Exchange Chromatography with Pulsed Amperometric Detection (HPAE-PAD), provided significantly better precision, allowing identification of structural isomers and simultaneous analysis of neutral and sialylated oligosaccharides.
